Tesofensine Capsules: What You Need to Know

Tesofensine is a phenyltropane-family drug that works as a serotonin-noradrenaline-dopamine reuptake inhibitor. It is being developed as a treatment for obesity, primarily as an appetite suppressant, but it may also increase resting energy expenditure and promote fat oxidation. Researchers believe that tesofensine modulates dopamine activity, which affects the pleasure sensation involved in food consumption.

How Tesofensine Works

Tesofensine capsules help suppress appetite and boost metabolic rate. Studies have shown tesofensine to be effective in treating obesity. In one study, participants taking tesofensine saw an overall 5-10% reduction in body weight, along with decreases in body fat, waist and hip circumference, and BMI. Tesofensine has also been found to have positive effects on cognition, memory, and learning. For these reasons, researchers are studying the drug for its potential use in treating depression and cognitive decline.

Clinical Trials

In a Phase II trial reported in The Lancet, tesofensine was found to produce significantly greater weight loss over a 6-month period than any currently available obesity drugs. Participants lost an average of 12.8 kg on the 1 mg dose, 11.3 kg on the 0.5 mg dose, and 6.7 kg on the 0.25 mg dose, compared to a 2.2 kg loss in the placebo group.

In the same Phase II trial reported in The Lancet, tesofensine was also found to significantly improve metabolic parameters, including insulin sensitivity and lipid profiles, compared to placebo.

Benefits of Tesofensine

Tesofensine has been found to have potential benefits beyond weight loss. Research suggests that it may also have positive effects on mood, leading to it being referred to as “the happy pill.”

Research suggests that tesofensine may also be helpful in treating depression and managing pain. Stress can decrease levels of BDNF (brain-derived neurotrophic factor), which is responsible for stimulating neuron growth and easing symptoms in those with depression and cognitive decline. While research is limited, some evidence suggests that tesofensine may help manage pain.

Side Effects

The safety profile of tesofensine is similar to that of currently approved medications for obesity treatment. The most commonly reported side effects in obese patients were dry mouth, headache, nausea, insomnia, diarrhea, and constipation.

Who Should Avoid Tesofensine

Generally, individuals who fall into the following categories should not take tesofensine:

  • Pregnant or breastfeeding women: The safety of tesofensine during pregnancy and breastfeeding has not been established, and it is recommended to avoid its use during these periods.

  • Individuals with cardiovascular conditions: Tesofensine can potentially increase heart rate and blood pressure. Individuals with a history of heart disease, heart rhythm disorders, uncontrolled hypertension (high blood pressure), or other cardiovascular conditions should avoid tesofensine.

  • People with a history of mental health disorders: Tesofensine may have effects on mood and brain function. Individuals with a history of mental health disorders, including depression, anxiety, or bipolar disorder, should exercise caution and discuss the potential risks with their healthcare provider.

  • Those taking certain medications: Tesofensine may interact with other medications, such as monoamine oxidase inhibitors (MAOIs), selective serotonin reuptake inhibitors (SSRIs), or other drugs that affect serotonin or dopamine levels. It is crucial to inform your healthcare provider about all medications you are currently taking to determine any potential interactions.

  • Individuals with glaucoma: Tesofensine can increase intraocular pressure and may worsen glaucoma symptoms. If you have glaucoma or a history of increased intraocular pressure, tesofensine should be avoided.
